80% RPMI 1640 + 20% FBS + 2 mM L-glutamine + 10 ng/ml muIL-3 or 10-20%
WEHI-3B supernatant split culture 1:2 twice a week; seed out at ca. 0.5 x 106
cells/ml; maintain at ca. 0.3-1.0 x 106 cells/ml; after thawing it is advisable to culture
the cells first in 24-well plates and to add 10 ng/ml mIL-3 or 10-20% WEHI-3B
supernatant daily; viability may be low after thawing at 37C with 5% CO2 maximum
density at about 1.2 x 106 cells/ml
